Senior RA/Principal RA/Scientist Molecular Biology and Cell Engineering

Dragonfly Therapeutics is seeking a highly motivated and experienced senior/principal research associate/scientist with a strong focus on molecular biology, mammalian cell culture, viral transduction, and flow cytometry to join the Biologics team in Copenhagen, (DK). The successful candidate will be a key member of the antibody discovery group responsible for design of plasmids and cloning, viral transduction and cell line development, CRISPR-mediated gene manipulation, and flow cytometric evaluation.


Responsibilities

  • Vector Design and cloning of plasmids for expression of recombinant proteins, antibodies and TriNKETs
  • Virus-mediated transduction and creation of isogenic cell lines expressing surface proteins
  • Cell-based screening and Flow cytometric evaluation and characterization of antibodies/TriNKETs
  • CRISPR/Cas9 mediated manipulation of cell lines for gene knock-in or knock-out
  • Molecular biology including sequence analysis, qPCR, RT-PCR, restriction digest and gel electrophoresis
  • Mammalian cell culture and high throughput protein production
  • Interface with CROs for sequence analysis, DNA scale-up, and cell line development
  • Generate, manage, analyze, and maintain critical data in a highly organized manner

Qualifications

  • B.S or M.S. with 5+ years of industry experience or Ph.D. in cell line development and characterization (title and salary commensurate with experience)
  • Extensive experience in vector design, Gibson, Golden Gate and restriction cloning, and creation of plasmids for expression of recombinant proteins and antibodies/TriNKETS
  • Proficiency in design and creation of retroviral/lentiviral plasmids, transduction of cell lines and primary cells
  • Hands-on experience in CRISPR/Cas9 mediated gene manipulation
  • Proficient in Flow Cytometric evaluation and characterization of cell lines for antibody binding and potency
  • Experience in extraction of immune cells from blood and their vitro characterization will be preferred
  • Experience in protein expression using Expi293 and ExpiCHO cells is a plus
  • Excellent aseptic skills, and mammalian cell culture experience is a must.
  • Hands-on experience with standard molecular biology skills including bacterial culture and transformation, qPCR, RT-PCR, DNA purification, restriction digestion, sequence analysis and vector construction.
  • Experience working with CROs for sequence analysis, DNA scale-up and cell line development
  • Efficient time management and ability to work on several projects simultaneously
  • Strong interpersonal skills and ability to effectively communicate with members of the multidisciplinary team
  • The ability to work collaboratively, provide strategic input, communicate clearly both written and orally is critical for success in this position.
  • Excellent record keeping abilities, to document analytical data generated in support of regulatory requirements, adherence to GDP, experience with ELN
